What this workflow helps you do
Measure distances, areas and angles directly on a scientific image while keeping each value connected to the geometry that produced it. The NotyMeasure workflow starts with calibration, moves through manual feature marking, and ends with a reviewable set of measurements and exports.
Use this workflow when
- Measuring feature length, spacing, diameter, area or angle on microscopy and other scientific images.
- Collecting repeated manual measurements when automatic segmentation is not appropriate or trustworthy.
- Keeping annotated geometry and numerical results together for later review.
What you need before you start
- An image with boundaries or landmarks that can be placed consistently.
- A verified calibration when physical units are required; otherwise results remain in pixels.
- A clear measurement rule, such as which edges, centerlines or endpoints will be used across repeats.
Step-by-step workflow
- Open the image in NotyMeasure and verify the attached calibration or accept pixel units intentionally.
- Choose the geometry that matches the question: line or path for length, a closed shape for area, or an angular tool for direction and intersection.
- Zoom to the feature and place the measurement points manually on the intended boundaries.
- Review the geometry and value in the Results area; correct or remove measurements whose point placement is inconsistent.
- Repeat the same measurement rule across the required features or fields of view.
- Review the result distribution or selected values, then export the annotated image and measurement data needed for your analysis.
What to review in the result
- Every retained value is linked to a visible, correctly placed geometry.
- All measurements being compared use the same calibration and unit.
- Outliers are checked against the image rather than removed only because they differ numerically.
- Repeated measurements follow the same boundary-placement rule.
Before you report or export
- Record whether values are calibrated physical measurements or pixel measurements.
- Keep the source image and calibration with exported tables or reports.
- For repeated measurements, document how fields and features were selected so the sampling can be understood.
Limitations and interpretation
- Manual measurement accuracy depends on resolution, boundary visibility, zoom and point placement.
- Calibration error propagates directly into converted physical lengths and areas.
- NotyMeasure does not automatically separate touching or overlapping particles; manual geometry remains the evidence for the reported value.
