Review Particle and Cell Segmentation

Choose a particle segmentation method and inspect object boundaries, touching objects and edge exclusions before accepting measurements.

Choose a suitable method

Start with the image contrast and object shape. Use an available watershed or manual approach, or choose Cellpose when an appropriate model and the NotyLab Local Engine are available.

Inspect the segmentation

  1. Open the image and confirm the scale if physical sizes are needed.
  2. Select a method and run segmentation.
  3. Inspect masks over the original image.
  4. Review touching, split and edge objects consistently.
  5. Check a representative subset before saving object measurements.

Keep comparison settings consistent

Record the method and selection rules used for each sample. A change in segmentation behavior can change the count and distribution even when the underlying image is unchanged.
